It has been established the mechanical loading have important role in regulating bone metastasis. However, it is unclear how cancer cell regulate bone cell mechaosensitivity, which plays a major role in determine bone quality.
Bone cells will be co-cultured with cancer cells under different loading conditions. Shear stress will be applied to bone cell only. Cancer cell released cytokines will be collected and used to treat bone cells with either loaded or non-loaded bone cells. Key mechanosensing related genes will be analyzed at both protein and mRNA level.
I will be conducting fluid flow loading experiment under the supervision of a PhD. student and will be responsible for collecting and analyzing experimental data.
